Biotin is widely used in biomarker and signal amplification techniques due to its unique chemical properties, especially in the biotin-streptavidin system, where streptavidin binds biotin with extremely high affinity. However, biotin is naturally present in biological samples and may be present in higher concentrations due to dietary intake, supplement use, etc. This can affect the results of biotin-labelled based assays, resulting in reduced sensitivity and increased likelihood of false negatives or false positives.

Biotin neutralising agents are designed to counteract this problem by neutralising and removing excess biotin from the sample, ensuring that the biotin-labelled assay system is not interfered with, and thus ensuring the accuracy and reliability of in vitro diagnostic results. Specific measures may include the use of anti-biotin antibodies, biotin-neutralising agents or other substances capable of competitively binding streptavidin to biotin, thereby restoring and maintaining the original sensitivity and specificity of the assay.

Application of the biotin-streptavidin system

The Biotin-SA (Biotin-Streptavidin) system is a conventional and broad-spectrum amplification system that takes advantage of the fact that one affinity molecule can specifically and rapidly bind to four biotin molecules to achieve multistage amplification. It is widely used in in vitro diagnostic reagents to enhance sensitivity.

Effects of biotin

Biotin is widely distributed in human blood and tissues and has a direct effect on the reagents using the Biotin-SA system, severely affecting the sensitivity and linearity of the reagents and leading to large deviations in the results.
