YODO ™ Blocking agents are a series of fully synthesized polymers with branched superhydrophilic groups and  bonding groups. The dense protective layer formed by the superhydrophilic group of this unique structure can significantly reduce non-specific adsorption, while the abundant bonding groups form stable and firm anchors on the surface to be closed.

YD-09, YD-11 and CE510 were used respectively in β2-MG Test Kit (Latex Enhanced Immuno-turbidimetry Method), the results shown excellent performance in improving the stability of the reagents, moreover, YD-09 and YD-11 help to enhance the sensitivity of the reagent obviously.

Usage & Dosage

  1. To 100mL of reagent solution that has been protein-coupled, 2mL of selected YODO ™ Blocking agent was added, and then mixed at 25℃ for 30 minutes. It is recommended to adjust the adding sequence and amount of blocking agent according to the actual application.
  2. The finished reagent is stored under suitable conditions, such as 2~ 8℃.

Storage conditions

Store at 2-8℃ is stable. Do not freeze it.

Eliminates undesirable effects of non-specific adsorption

The binding groups of YODO™ Blocking agent form stable and firm anchors on the surface through chemical covalent, in the meantime, the dense protective layer formed by the superhydrophilic groups of YODO™ Blocking agent can significantly reduce non-specific adsorption.

YODO™ Blocking agent is fully chemically synthesized, which has no biosafety risk compared to BSA, as it contains no biological components. In addition, YODO™ Blocking agent has no difference in lots, high purity and consistent quality, which contributes to the reliability of experimental results. It has a unique spatial structure that forms a dense protective layer and reduces non-specific adsorption, thus reducing matrix interference and improving sensitivity and stability.

The application of YODO™ Blocking agent in immunodiagnostic reagents can make up for the shortcomings of BSA as a blocker, which can be immobilised on the surface of the solid phase by covalent binding and/or physical adsorption, and can also be reversibly bound to biological macromolecules, such as proteins and nucleic acids, to effectively protect the stability of these macromolecules, effectively reduce non-specific adsorption, increase the stability and sensitivity of the reagents, and improve the quality of the test results.

Non-specific adsorption (NSB) refers to the phenomenon of non-covalent bond adsorption without selectivity, which is ubiquitous, especially in the IVD field, where non-specific adsorption of the reagent vial to the contents and the substance to be tested may occur between or among themselves.

NSB leads to a decrease in the sensitivity and specificity of the reagents, an increase in background noise, and possible false-positive results, affecting the accuracy and reliability of the test. NSB is very common in the field of IVD, which may lead to a reduction in the effective binding area of the biosensor, affecting the detection sensitivity, and even causing agglutination or inflammatory reactions on the surface of the material, and the use of a suitable blocking agent is one of the measures to solve this problem.

The main causes of non-specific adsorption include differences in surface energy due to heterogeneity, hydrophobic interactions, electrostatic interactions and hydrogen bonding. These forces lead to non-specific adsorption between substances of different compositions.

When selecting the blocking agent, the specific conditions and needs of the experiment should be considered to the specific application scenarios, firstly determine the type of groups to be blocked, for example, YD-03, YD-05, YD-09, YD-11, YD-13, YD-14, YD-15 for carboxylic acid, sulfonyl, epoxy, chloromethyl, and YD-17 and YD-18 for the colloidal gold or other metal particles.

How does the YODO™ Blocking agent couple to the activated carboxyl groups on the surface of the microsphere?

Chemical covalent coupling is achieved through the formation of stable covalent bonds between the bondable groups in the sealant and the functional groups to be sealed (e.g. carboxyl groups). Physical adsorption, on the other hand, utilises non-covalent forces such as electrostatic interaction, hydrogen bonding, van der Waals forces, etc. between the molecules of the sealant and the substance to be sealed.

YODO™ Blocking agent inhibits NSB of proteins by forming a dense hydrophilic layer on the surface of the microspheres, which helps to maintain the correct stereo conformation of the antibody, resulting in enhanced signals and desirable detection results.
